By using negative selection immunomagnetic method’s Peripheral blood primary human monocytes could be isolated from mononuclear cells. Since the Monocytes are isolated with immunomagnetic method’s they can be directly implemented in any downstream application. Peripheral blood was collected using one of the following anticoagulants: ACDA, ACD, CPD, CP2D, or CPDA.

Cells were obtained using approved consent forms, Review Board and protocols.

Cell Type: Myeloid Cells and Monocytes

Cryopreserved Monocytes can differentiate into Dendritic Cells and Secrete p70 and IL-23. Upon Activation
Monocytes freshly isolated from a Leuko Pak (leukopak is an enriched leukapheresis product collected from normal peripheral blood) using cryopreserved Monocytes were cultured for 6 days in RPMI 1640 Medium with 10% FBS, 0.1 mM MEM Non-Essential Amino Acid Solution, 2mM L-Glutamine, 1mM Sodium Pyruvate and 50µM β-mercaptoethanol. Human Recombinant IL-4 and Human Recombinant GM-CSF were added on days 1, 3 and 6 to differentiate monocytes into DCs. Only activation with Human Recombinant IFN-γ led to secretion of (A) IL-12 (p70) and (B) IL-23, (measured using the Human p70 and IL-23 ELISA Kit).
